Introduction


EGFR pharmDx Kit is a qualitative immunohistochemical assay. It includes all reagents necessary to identify the expression of epidermal growth factor receptor (EGFR) protein on the surface of normal and neoplastic cells. The kit will provide reproducible results with its’s standard protocol. EGFR is indicated as an aid in identifying colorectal cancer patients eligible for treatment with Erbitux® (cetuximab) or Vectibix™ (panitumumab).


The EGFR pharmDx™ Kit Includes:

Procedure


The kit utilizes a simple two-step staining procedure and is suitable for formalin-fixed, paraffin-embedded specimens. Results can be available within 1 day, giving clinicians EGFR expression levels in a quick and reliable manner. This kit employs a ready-to-use visualization reagent based on dextran technology. A common dextran polymer backbone links both secondary goat anti-mouse antibody molecules and horseradish peroxidase molecules, thus eliminating the need for the sequential application of link antibody and peroxidase conjugate. Further, the addition of chromogen results in the formation of a visible reaction product at the antigen site. Later, the specimen may be counterstained and coverslipped.


Results Interpretation


Results are interpreted using a light microscope. Control slides containing two formalin-fixed, paraffin-embedded human cell lines with staining intensity scores of 2+ and 0 are provided for validation of the kit reagent performance.
