The Quintessential Accessory to NGS for CNV Screening: SALSA digital MLPA

A variant of multiplex polymerase chain reaction (PCR) is the multiplex ligation-dependent probe amplification (MLPA). In this technique only a single primer probe is used to amplify multiple targets. Using MLPA, copy number variations (CNV) are detected at a molecular level, and the analysis is done using the specific software program called Coffalyser.NET.
Selective Adaptor Ligation, Selective Amplification (SALSA)digitalMLPA runs on the same principle as a conventional MLPA but has a greater advantage of detecting many more targets. It’s the amalgamation of MLPA technology and Next-generation sequencing (NGS) for CNV detection for a larger number of targets.
With reduced costs and turnaround times, digitalMLPA can detect up to 1000DNA sequences in a simple multiplex PCR-based reaction. The PCR products or amplicons that are generated in the digitalMLPA can be quantified using Illumina NGS platforms. The read numbers of each digitalMLPA probe amplicon is determined by Sequencing.
The aim of digitalMLPA is to increase the efficiency of CNV screening. D001 Hereditary Cancer Panel 1, the first SALSA digitalMLPA panel by MRC-Holland, has greater gene coverage than that of ten conventional MLPA assays thus streamlining the oncogenetic screening process.
CNVs in 28 clinically relevant genes and six specific point mutations are detected by the digitalMLPA probemix D001. The hereditary predisposition to one or more of the following cancer types: breast, colorectal, gastric, ovarian, endometrial, prostrate, melanoma, and pancreatic depends on the mutation statuses of the aforementioned genes.
A single chip can be used to run digitalMLPA reactions and NGS libraries therefore making digitalMLPA the ideal time-saving accompaniment to NGS for high-level CNV screening.
